Anfangs der 20er Jahre verliess Louise Fazenda die Company von Mack Sennett und setzte ihre Filmkarriere mit Spielfilmen fort, in denen sie ebenfalls ihr komödiantisches Talent beweisen konnte.

Der Übergang zum Tonfilm bereitete ihr keine Probleme und setzte ihre Laufbahn mit Filmen wie "Wide Open" (30), "Viennese Nights" (30), "Gun Smoke" (31), "Racing Youth" (32), "Alice in Wonderland" (33), "Caravan" (34), "Bad Boy" (35), "The Widow from Monte Carlo" (35), "The Road Back" (37), "Ever Since Eve" (37) und "The Old Maid" (39).

1939 zog sie sich vom Filmgeschäft zurück und setzte sich für humanitäre Bedürfnisse ein und wurde eine begeistere Kunstsammlerin.

Louise Fazenda war in zweiter Ehe mit dem berühmten Produzenten Hal B. Wallis verheiratet und man scherzte, dass Wallis "the Prisoner of Fazenda" war (in Anlehnung an das berühmte Stück "Der Gefangene von Zenda").
