Dort wurde sie für den Film entdeckt und aus dem Ausflug wurde ein ständiger Aufenthalt in Hollywood. Ihre Familie zog ebenfalls nach Kalifornien - ihre beiden Schwestern Diana Kane und Connie Lewis wurden ebenfalls Schauspielerinnen.

Lois Wilson erhielt erste Nebenrollen und spielte in einer Vielzahl von Filmen mit wie "The Girl of the Night" (15), "The Hypocrite" (15) - wo sie zusätzlich auch die Regie führte, "Hulda the Silent" (16), "The Silent Battle" (16), "The Beckoning Trail" (16), "Black Evidence" (17), "Alimony" (17), "A Burglar for a Night" (18), "The Bells" (18), "Three X Gordon" (18), "The End of the Game" (19) und "The Price Woman Pays" (19).

1919 erhielt sie von Paramount einen Vertrag und konnte in den 20er Jahren in namhaften Produktionen wie "The City of Maks" (20), "Miss Lulu Bett" (21), "Manslaughter" (22), "The Covered Wagon" (23), ""Bella Donna" (23) und "The Man Who Fights Alone" (24).

Ab Mitte der 20er Jahre war Lois Wilson ein Star und spielte mit den berühmtesten Filmpartnern zusammen auf der Leinwand. Unter anderem agierte sie an der Seite des grössten Stars der Stummfilmzeit im Film "Monsieur Beaucaire" (24) - Rudolph Valentino.

Weitere namhafte Produktionen in den 20er Jahren waren "Icebound" (24), "Bluebeard's Seven Wives" (26), "The Great Gatsby" (26), "New York" (27), "Ransom" (28), "Conquest" (28) und "The Gamblers" (29).

Der Tonfilm bot ihr weiterhin viele interessante Rollen an, meist jedoch Nebenrollen. In den 30er Jahren entstanden u.a. die Filme "Temptation" (30), "The Expert" (32), "Divorce in the Family" (32), "Secrets of Wu Sin" (32), "The Crash" (32), "Female" (33), "Ticket to a Crime" (34), "Life Returns" (35) und "Bad Little Angel" (39).
Nach den Filmen "Nobody's Children" (40) und "For Beauty's Sake" (41) zog sie sich bis auf wenige Ausnahmen vom Film zurück. Ihre letzten schauspielerirschen Tätigkeiten waren für das Fernsehen - die Serien "The Guiding Light" (54) und "The Secret Storm" (54).
